What is MGF?
Mechano Growth Factor (MGF) is a splice variant of IGF-1 that is produced locally in muscle tissue in response to mechanical loading and damage. It was identified in the late 1990s by researchers studying why exercised muscle expressed a different form of IGF-1 than systemic IGF-1.
The distinct feature of MGF is its E-domain, which appears to have biological activity independent of the IGF-1 receptor. Research suggests this E-domain plays a primary role in activating muscle satellite cells — the resident stem cells of muscle tissue responsible for repair and growth. Native MGF has an extremely short half-life of only a few minutes, which led to the development of PEG-MGF, a PEGylated version with hours-long activity.
Research dosing typically cites 100-200 mcg locally injected post-training, with the rationale being that MGF should be present during the window when satellite cells are being activated by mechanical loading. The clinical research base is thin — most published data comes from in vitro and animal studies, with limited human work.
MGF is sometimes positioned as superior to IGF-1 LR3 for muscle hypertrophy because of the satellite cell activation mechanism. The honest assessment is that the research isn't strong enough to make that claim with confidence — both peptides have potential mechanisms that look interesting on paper, and both have limited human data.
